PURPOSE OF POSITION: To learn and perform the state-of-the-art genome editing techniques and to generate gene-edited cell lines as well as animals. To learn and perform specialized transgenic techniques, including sperm/embryo cryopreservation, embryo transfer, microinjection, IVF and so forth. To provide reliable and high-quality service to both internal and external investigators with the highest standard of work ethic. Work collaboratively with others, as a team member, to develop positive working relationships and to achieve goals. We are looking for an individual who pays attention to detail and is motivated in learning new techniques. Experience in molecular cloning, cell culture, and animal handling is preferred. Experience in embryo manipulation is a plus.

Responsibilities

Molecular Biology and Cell Culture (main responsibility): Perform basic molecular biology experiments, including plasmid cloning and amplification, electrophoresis, cell culture and transfection, maintain and target embryonic stem (ES) cells and cell lines, PCR, enzyme digestion, TA cloning and sequencing, and so forth. Perform CRISPR/Cas technology, genome-editing project design and genotyping, and prepare final reports for investigators. Apply standard protocols to specific experiments, improve current methods and evaluate innovative techniques. Maintain accurate and detailed records of all laboratory work and experiments.

Superovulation and Embryo Isolation: Perform superovulation in female animals by hormone injection. Isolate fertilized eggs or 4-8 cell embryos/blastocysts from oviducts and uteri. Perform pronuclear and cytoplasmic microinjection to generate transgenic and genetically modified animals. Proficient in microinjection of 8-cell embryos/blastocysts with pluripotent ES cells to generate chimeric animals. Work on weekends and holidays when needed.

Cryopreservation Techniques: Perform techniques for storage of transgenic mouse lines, including embryo stepwise cryopreservation, embryo thaw, sperm cryopreservation, in vitro fertilization and mouse recovery.

Animal Husbandry Maintenance: Assist all aspects of animal husbandry for the Transgenic Animal and Genome Editing Core Facility when needed. Maintain an accurate census of the mouse colony, anticipate transgenic work load and order mice only when appropriate, observe daily for health status, hormone administration, animal mating, appropriate care and attention to pregnant recipient females and subsequently to potential founder animals. Manage sentinel program, accurately mark animals for identification, perform tail biopsies, accurately identify positive transgenic founders, appropriate monitoring of animals post-surgically, and timely transfer of "sentinel checked" transgenic animals to individual researchers.

Contribute to and promote a positive and professional work environment/atmosphere. Maintain currency in field through continuing education, literature and seminars. Implement this knowledge into the current protocols. Participate in improving organizational performance through recommending areas or approaches for improvements activities, performing new procedures, collecting data and providing input to department discussions.
